基因组DNA (Genomic DNA)
基因组DNA是一种确定分子检出限的理想工具,但若要更加有效,则需要在细胞系中提取出精确拷贝数及等位基因的gDNA,而这一切在患者细胞中并不容易获得。HDx参考标准品因其具有高度特色,其天然可用于生产DNA的细胞系成为理想的解决方案。
DNA参考标准品
Horizon的DNA标准品源自人工突变体(50%突变体)和匹配“正常”(0%突变体)的细胞系。突变细胞系都是杂合子,在大多数情况下,这意味存在着单个突变等位基因和单个正常等位基因。当拷贝数为2N时,100%的突变型DNA样品表明提取DNA的细胞中100%含有该突变,但由于该细胞的两个拷贝数中实际只有一个含有该突变,实际上该样本的等位基因突变频率为50%。
具有3N拷贝数和单个突变等位基因的基因型含有33%的等位基因突变频率。可以将突变体和“正常”DNA精确混合以产生具有特定等位基因频率的标准品。
